Idiopathic pulmonary fibrosis (IPF) is a progressing chronic and fibrotic lung response with poor prognosis. To study the underlying molecular mechanisms of IPF, the rat bleomycin model is commonly used. Intratracheal application of bleomycin is known to induce inflammatory and fibrotic processes in the lung, e.g. infiltratin of inflammatory cells and collagen deposition. Recently, the PDE4B inhibitor BI 1015550 has been reported to prevent a decrease in lung function in patients with IPF. Here we examined the effect of PDE4 inhibitor BI 1015550 in the rat bleomycin model on transcriptional level by RNASeq. Overall design: Here we asses the effect of therapeutic PDE4B inhibition in the rat Bleomycin model of pulmonary fibrosis via RNASeq on the lung transcriptome. The experiment consisted of control animals (n=6), Bleomycin treatment (1mg per kg in 0.9 percent NaCl aequeous solution) on day 0 (n=5), treatment with PDE4B inbibitor (BI 1015550) between day 10 and day 20 twice daily (n=6). The material collection took place on day 19 for the control group and on day 20 for treated animals.
