Saccharomyces cervisiae Transcriptome upon misfolded protein stress in Cytosol

Study to identify stress response pathways upon single misfolded protein stress in the cytosol. Single genome integration of Wild type and misfolded protein gene of NAT (Nourseothricin acetyl transferase) in URA locus. Cultures grown in rich media (YPD) at 30 degree celsius till exponential phase and total RNA was isolated. From total RNA mRNA enrichment was done using Dynabeads mRNA DIRECT Purification Kit. Enriched mRNA was sequenced on ION torrent platform after cDNA conversion. Control strain is strain with WT-NAT integration. Others are misfolded mutants of NAT depending upon severity of mutants (TS22 being most severe, TS15 moderate mutant and TS5 as mild mutant. All were done in three biological replicates.
