This dataset contains transcriptomic data from human THP-1 macrophages with and without serglycin (SRGN) gene knockout, under basal (M0) and pro-inflammatory (M1) polarization conditions. Serglycin is a hematopoietic proteoglycan known to regulate the storage and secretion of inflammatory mediators in immune cells. This study investigates the role of serglycin in macrophage polarization, inflammatory signaling, and metabolic reprogramming.RNA-sequencing was performed on wild-type and CRISPR/Cas9-mediated SRGN knockout THP-1 macrophages. Cells were differentiated into macrophage-like M0 state using PMA and subsequently polarized to an M1 state with lipopolysaccharide (LPS) and interferon-gamma (IFN-g). The dataset includes six biological replicates for each condition (wild-type M0, wild-type M1, SRGN knockout M0, SRGN knockout M1).The findings reveal that serglycin deficiency modulates inflammatory gene expression, cytokine secretion, vesicle biogenesis, phagocytic capacity, and mitochondrial respiration in macrophages. Differential expression analysis highlights pathways associated with TNF-a signaling, interferon responses, glycolysis, and reactive oxygen species (ROS) production. This dataset provides insights into the molecular mechanisms underlying serglycin's role in macrophage function and its potential implications in inflammation and metabolic regulation.
