We establish that subtelomeric regions together with some other functional elements like transposons are consistently under-replicated in metaphase. Inhibition of CDK activity, in fact, results in accomplishing of DNA replication in those regions. Overall design: To answer the question, we sequenced samples arrest in G1-phase and M-phase (by using a strain with methionine-sensitiver promoter, placed in front of CDC20). Additionally, we sequenced samples arrested in M-phase with additional inhibition of CDK activity
