Analyzing the regulatory effect of aqueous extract from Traditional Chinese Medicine Wuyi on the gastric mucosal microbiota in A4gnt gene knockout (A4gnt KO) mice. The aqueous extract of Wuyi was prepared using the decoction method. SPF-grade C57BL/6N mice aged 6-8 weeks were selected, including wild-type (WT group, n=10) and A4gnt KO mice (n=20). The A4gnt KO mice were randomly divided into the aqueous extract of Wuyi intervention group (KOWY group, n=10) and the standard diet control group (KO group, n=10). After 30 days of continuous gavage administration, mice were raised conventionally until 30 weeks of age before euthanasia. Gastric mucosal tissues were collected for histopathological HE staining and 16S rRNA gene sequencing analysis of the microbiota. An optimal microbial genus combination model distinguishing KO and KOWY groups was constructed based on AUC-RF algorithm. PICRUSt2 was employed to predict KEGG functional pathways of the microbiota.
