Presence of M. globosa DNA within pancreatic FFPEs
收藏资源简介:
This record contains the final data used to assess the presence of Malassezia globosa in pancreatic formalin-fixed paraffin-embedded (FFPE) tissue. It combines quantitative PCR (qPCR) results and fluorescence in situ hybridization (FISH) counts with RNA/DNA sequencing–based microbial profiles and accompanying metadata. The dataset supports the associated manuscript and allows readers to inspect the underlying Cq values, FISH counts, microbial abundances, and sample-level annotations. Contents qPCR and FISH TaqMan qPCR M. globosa in pancreatic FFPEs.xlsx – Cleaned per-sample qPCR table for M. globosa (target) and human beta-globin/HBB (reference/quality control), including FISH counts in column H. “Non-congruent” cases were repeated in the lab; the table retains only interpretable and consistent results. README_qPCR_min_EN.txt – Short README explaining the qPCR/FISH dataset, column structure, and how repeated runs were handled. Sequencing RNA_DNA_Sequencing_MicrobialCounts.tsv – Microbial count table derived from RNA/DNA sequencing of pancreatic FFPE tissue, reported per sample. RNA_DNA_Sequencing_Metadata.tsv – Sample-level metadata corresponding to the microbial count table (see file header for all variables). RNA_DNA_Sequencing_Unmapped_Fastq.zip – Compressed archive containing FASTQ files with unmapped reads from the RNA/DNA sequencing runs (see file naming inside the archive). Dictionaries simple_data_dictionary_EN.csv – Simple data dictionary listing column names, inferred data types, and brief descriptions for the main tabular files. Workbook info qPCR workbook sheets: Tabelle1 Main sheet used for the manuscript: Tabelle1 How to link the files Sample identifiers can be used to link qPCR/FISH results, microbial counts, and metadata across files (see README and column headers for details). For the full analytical workflow, quality-control steps, and statistical analyses, see the associated manuscript. Methods (plain language) qPCR (TaqMan): Detection of M. globosa (target of interest) and HBB (human quality/quantity control) on pancreatic FFPE material. Lower Cq values indicate more target; “No Cq” means the target was not detected. Non-congruent technical replicates were re-run, and only interpretable, consistent results are included in the final table. FISH (fluorescence in situ hybridization): Probe-based visualization and counting of signals per specimen; FISH counts are reported in column H of the qPCR Excel file (see manuscript for probe design and scoring criteria). RNA/DNA sequencing: Library preparation, sequencing platform, read processing, and taxonomic assignment are described in the accompanying manuscript. This record provides the final per-sample microbial counts, the matching metadata, and the unmapped read FASTQs to enable re-analysis.
