This BioProject includes several RNA sequencing datasets associated with the following experiments. (i) Small RNA-seq dataset of P0 and F1 N2, app-1(ths372), and dpf-3(ths1047) animals exposed to oma-1 RNAi: This dataset contains total small RNA sequencing data from Caenorhabditis elegans wild-type N2, app-1(ths372), and dpf-3(ths1047) animals exposed to oma-1 double-stranded RNA (dsRNA). Total small RNA libraries were prepared from the directly exposed parental generation (P0) as well as from their progeny (F1), followed by high-throughput sequencing. (ii) Small RNA-seq analysis of N2, app-1(ths372), dpf-3(ths1047), and app-1(ths372); dpf-3(ths1047): This dataset comprises total small RNA sequencing data from C. elegans wild-type N2 and mutant strains app-1(ths372), dpf-3(ths1047), and the double mutant app-1(ths372);dpf-3(ths1047). Total small RNA libraries were prepared from young adult worm populations reared at 20 degree and sequenced to profile small RNA expression and compare small RNA composition across genotypes. (iii) Small RNA-seq datasets of RNAs immunoprecipitated with WAGO-1, WAGO-3, and WAGO-4 (WAGO RIP-seq): Animals expressing WAGO-1::FLAG, WAGO-3::FLAG, or WAGO-4::FLAG were exposed to either control or oma-1 RNAi treatment. Each WAGO protein was immunoprecipitated from lysates prepared at the young adult stage using an anti-FLAG antibody to enrich for WAGO-1, WAGO-3, and WAGO-4 associated RNAs. The co-purified RNAs were then subjected to high-throughput sequencing to profile small RNAs bound to WAGO-1, WAGO-3, or WAGO-4.
