This study aims to systematically review the current technological landscape and future potential of mitochondrial function restoration strategies for extending lifespan and healthspan. Mitochondria play a central role in cellular energy metabolism, oxidative stress regulation, inflammatory signaling, and the maintenance of cellular homeostasis. Age-related mitochondrial dysfunction is widely recognized as a key driver of aging processes and multiple age-associated diseases. This research categorizes major intervention approaches, including mitophagy activation (e.g., Urolithin A), inner membrane–stabilizing peptides (Elamipretide/SS-31), mitochondria-targeted antioxidants (e.g., MitoQ), electron transport chain cofactors (CoQ10), metabolic and biogenesis pathway modulation (PGC-1α/AMPK), NAD⁺-linked mitochondrial support strategies, and emerging cell- or organelle-based therapeutic approaches. In addition, the study provides a structured framework of standardized evaluation methods, incorporating functional respiration assays (OCR/ECAR), mitophagy flux markers, ROS indicators, and mtDNA-based biomarkers, thereby improving reproducibility and cross-study comparability. Overall, mitochondrial restoration technologies show expanding potential applications across diverse areas such as muscle decline, immune aging, metabolic disorders, and neurodegenerative diseases. With further advancements in biomarker refinement and combination strategies, these approaches are expected to contribute significantly to the development of next-generation platforms for healthy longevity research and therapeutic evaluation.
