Plants of P. trichocarpa were grown in a greenhouse under 16 h light/8 h dark conditions photoperiod, with light provided by cool white fluorescent lights (250 μmol m-2 s-1 PPFD). For stress treatment, we took fifteen plants obtained from one genotype, using three for chilling stress (4°C, 6 h, 250 μmol m-2 s-1 PPFD), three for heat stress (42°C, 6 h, 250 μmol m-2 s-1 PPFD), three were exposed to drought treatment (soil water-content level ;RSMC 20%), three were exposed to 150 mM NaCl, 30% polyethylene glycol (PEG) 6000 for 6 h, the other three as controls under light (25℃, 6 h, 250 μmol m-2 s-1 PPFD). Leaves for RNA extraction were collected from P. trichocarpa with or without treatment. The RNeasy Plant mini kit (Qiagen, Hilden, Germany) and Super-Script First-Strand Synthesis system (Invitrogen) were used for total RNA extraction and cDNA synthesis, respectively. Strand-specific was performed on an Illumina Hiseq2005.
