Huntington's disease (HD) is caused by a CAG repeat expansion in the huntingtin (HTT) gene. The CRISPR-Cas9 nuclease causes double-strand breaks (DSBs) in the targeted DNA, which can be toxic, whereas CRISPR interference (CRISPRi) using dead Cas9 (dCas9) suppresses the expression of a specific gene without inducing DSBs.To compare the effects of Cas9-sgRNA and dCas9-sgRNA on the whole striatum transcriptome, RNA samples were prepared from the brain region at 12-13 weeks of age.
