Balbc J mouse received vehicle (solvent) or OGG1 inhibitor 1, 4, 8, 12, 24, 28 , 32, 36, 40 and 44h after RSV infection via intraperitoneal route. RSV (10^6 PFU) was installed into lungs via intranasal route in 60 microliters of solvent). One, 2, 3, 6, 12, 24, 48 and 72 hour after RSV challenge mice were sacrificed, lungs were removed and RNAs were isolated from individual lungs (n=6) After reverse transcription cDNAs were pooled and changes in gene expression was determined by using SABiosciences Mouse Inflammatory Cytokines & Receptors PCR Array (PAMM-011ZD) Total pooled (n=6) cDNAs generated Superscript® III First Strand Synthesis System (Invitrogen), mixed with equal amounts of 2X SYBR Green Supermix (Qiagen) and 20 μl of reaction mixture was added to each well of the PAM-011ZD array. The reaction was evaluated using an ABI PRISM® 7000 Sequence Detection System using recommended settings by SABiosciences.
