RNA-seq of depletion Mina53 in HCT116 p53+/+ cells and HCT116 p53-/- cells

We report the signaling pathway of Mina53 by depleting Mina53 in HCT116 p53+/+ cells and HCT116 p53-/- cells based on RNA-sequencing. By using magnetic oligo(dT) beads to enrich the total RNA with poly A, and then breaking the RNA to fragments about 300bp, these fragments are used as models to get cDNA by using primers and reverse transcriptase, and then the second cDNA model. After the library was constructed, we use next-generation sequencing (NGS) based on Illumina HiSeq sequencing platform to detect the DNA sequence. After filtrating the raw data, the clean data was blast to the human genome sequence.
