RNA microarrays technology was used to compare hESC-derived cell populations to undifferentiated hESC and to human EL cell populations. Microarrays confirmed the hematoendothelial, endothelial and hematopoietic identity of hPSC-derived CD144+-EBs, BCs and ECs, respectively, and the similarities between hESC-derived cell populations and human EL equivalent populations. hPSC-EC, hPSC-CD144+EB, hPSC-BC and hPSC CD144+CD45+BC were derived from undifferentiated hESC (H1). Endo 144+45-34+, Endo-Hemato 144+45low34+, Hemato 144-45low34+, Hemato 144-45high34- cell populations were freshly isolated from human EL. Undifferentiated hESC (H1), hESC-derived cell populations and EL cell populations were lysed and RNA was extracted of each sample. RNA was quantified and qualified after purification and same quantity of RNA was used for each sample for microarray analyses.
