Tcf1-CTCF cooperativity shapes genomic architecture to promote CD8+ T cell homeostasis [Hi-C]

An intricate interplay among Tcf1/Lef1, CTCF and Stat5 that orchestrates genomic architecture to coordinately promote CD8+ T cell homeostatic proliferation Splenocytes were harvested from hCD2-Cre, Rosa26-lsl-GFP, Tcf1/Lef1 wild-type or double floxed mice, and sorted for GFP+TCRbeta+ naïve CD8+ T cells. The sorted cells were stimulated with IL-7 and IL-15 for 72 hrs, followed by crosslinking, enzyme digestion, proximity ligation, and then HiC sequencing.
