In this study we describe the immunosupressive role of Tim3+ tumor cells during eraly stages of metastasis. We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of immune cells subsets altered by Tim3 expression in the tumor cells. The aim was to determine the immune status of immunosuppressive and cytotoxic populations during the dynamic process of the metastatic colonization in the liver Intracardiac injection of 4TO7-Ctrl and Tim3-KD cells into Balb/c was performed. At different timepoints (micrometastasis, day 3) and (macrometastasis, day 12), immune cells were isolated from a liver section (5x5mm). Also adjacent tissue of the liver free of tumor was isolated. According to CD45 signal by fluorescent-activated cell sorting, cells were analyzed using scRNA-seq.
