Amplicon metagenome analysis to describe biofilm microbial communities from cobbles and sediment samples taken from an intermittent third order stream. Raw sequences from 16S rRNA amplicon sequencing (hiper-variable V4 region). The present data set consists of samples collected during the hydrological year 2019-2020, in spring (28 March), summer (6 June), fall (28 November), and winter (20 February) in Fuirosos (41.684583, 2.579444), an intermittent third order stream located within the Montnegre-Corredor Natural Park (Catalonia, NE Spain). For each season, samples were collected along a 100 m reach, divided longitudinally into 7 sections. In each section, two biofilm streambed sediment samples (i.e., epipsammon, 0-3 cm depth) were collected. Surface sediment samples (20-30 g each) were carefully transferred to 50 mL falcon tubes with 5 mL of lysis buffer (40 mM EDTA; 50 mM Tris pH 8.3; 0.75 M sucrose). In addition, we collected two cobbles (one located in a run zone and one in a riffle zone) to obtain epilithon samples at stations 1, 3, 5, and 7. In the laboratory, we extracted the epilithon from each cobble by scraping with a sterilized metal brush and washing them with ringer's solution. An aliquot of the sludge was filtered through a 0.2 micro-meters polycarbonate membrane and frozen at -20C degrees until subsequent microbial DNA extractions. The results were used to characterize the microbial community structure and to compare possible differences between seasons.
