Lactate dehydrogenase (LDH) and malate dehydrogenase (MDH) are homotetrameric enzymes forming a superfamily with a similar fold. Their biological activity is directly related to the tetrameric assembly and associated dynamics. Moreover, LDH are allosterically regulated while MDH not. We will used pressure perturbation to explore intrinsic rigidity/flexibility of both enzymes. The comparison of their pressure-response will allow us to identify the determinants of LDH allosteric regulation and the dynamics associated with providing new information on LDH/MDH biological activity.
