We have used single-nucleus (sn) RNA sequencing through a 10x Genomics platform to examine the impact of overexpressing CCN1 in astrocytes during the critical period. We transcriptionally profiled all cell types in the visual cortex after bilateral injection AAV-CCN1 or AAV-tdTomato as a control. Overall design: 14 days after AAV injection into the visual cortex bilaterally, the visual cortices were dissected out, nuclei were extracted and FACS was performed on Hoechst-labeled nuclei. 2 visual cortices per mouse were dissected and 2 mice were pooled per sample. 1 tdT sample and 1 CCN1 sample were considered 1 replicate. Replicates were sequenced at different times but samples were processed the same way.
