Chromosome-level assembly of Z. tritici progeny

The isolates result from the stablishment of a pedigree based on experimental crossing. The first sexual cross of the isolates 1A5 with 1E4 (PRJEB15648 and PRJEB20900 respectively) is described by (Lendenmann et al, 2015 G3). One of the 341 haploid ascospore from this progeny was selected for further sexual crosses. This isolate (M1_A66.2) was subjected to a backcross with the 1A5 parent following the protocol described by Kema et al. (1996) (2nd round of meiosis). Briefly, spores of the parental pair were sprayed on wheat plants and incubated until the development of pseudothecia. Ascospores ejected from those same pseudothecia were isolated on water agar plates. The germinating ascospores were transferred for clonal propagation and further DNA extraction. Similarly, one isolate from the resulting progeny was backcrossed again with 1E4 parent (3rd round of meiosis). Two isolates (M3_19 and M3_45) resulting from this cross were further backcrossed with 1A5 and 1E4 parent respectively (4th round of meiosis).
