Development of an RNA-based RT-LAMP assay for rapid detection of Brettanomyces bruxellensis in wine

This dataset contains experimental data related to the development and validation of an RNA-based RT-LAMP assay for the detection of Brettanomyces bruxellensis in wine. The study focuses on the amplification of the 26S ribosomal RNA (rRNA) as a marker for metabolically active cells. The dataset includes RT-LAMP amplification results obtained from serial dilutions of B. bruxellensis (ranging from 10⁷ to 10¹ cells/mL), as well as data from wine samples used for method validation. Ct values were used to generate standard calibration curves, enabling quantitative estimation of cell concentrations in unknown samples. RNA extraction was performed using commercial kits, including the YeaStar™ RNA Kit (Zymo Research) and the E.Z.N.A.® Total RNA Kit (Omega Bio-Tek), following the manufacturers’ protocols. RT-LAMP amplification was carried out using two different commercial mastermixes: the RT Isothermal Mastermix (OptiGene) and the Saphir RT-LAMP Kit (Jena Bioscience), in order to evaluate assay performance under different conditions. The dataset also includes comparative analyses with other detection techniques, such as DNA-based PCR assays and flow cytometry, used as reference methods to assess the reliability and sensitivity of the RT-LAMP approach. These data support the development of a rapid, sensitive, and cost-effective method for the detection of Brettanomyces bruxellensis in wine, suitable for potential application in routine wine quality control.
