Adenylate cyclase signaling pathway is suggested to be a key regulator of immune system functions. However, specific effects of cyclic adenosine monophosphate on T helper cell differentiation and functions are unclear. Involvement of cAMP in Th cell differentiation program, in particular development of Th1, Th2, and Th17 subsets, was evaluated employing forskolin (FSK), a labdane diterpene well known as AC activator. We performed scRNA sequencing to understand the distinct effect of forskolin on Th1, Th2 and Th17 differentiation program. We included following samples: non-treated control (NT ctrl, resting cells), activated control (ACT - anti-CD3/anti-CD28 stimulated cells), cells differentiated into distinct subsets: Th1, Th2, TH17, and these subsets treated with 10 µM forskolin: Th1+FSK10, Th2+FSK10, Th17+FSK10.
