This experiment allowed us to quantify the transfer efficiency of the TAPs by bacterial conjugation. Conjugation was performed using the donor E. coli MG1655 / F-Tn10 and either the TAP-Cas9-nsp or TAP-Cas9-lacZ2 mobilizable plasmids carrying the sfGFP green fluorescent reporter mixed with a lac+ recipient strain that produces the red fluorescent histone-like protein HU-mCherry. Transconjugants are identified by the production of both green and red fluorescence.
