The effect of the SASP upon liver sinusoidal endothelial cells

Primary human liver sinusoidal endothelial cells from 7 explanted livers were incubated in conditioned media from IMR90 cells undergoing several forms of senescence Overall design: Conditioned media was generated from IMR90 cells transduced with ER:RAS (4OHT-inducible HRAS^G12V) and XIH-N1ICD (Doxycyline-inducible NOTCH1 intracellular domain) treated with 4OHT or doxycycline or both, leading to RAS-induced senescence (RIS) or NOTCH-induced senescence (NIS) or NOTCH/RAS-induced senescence (N+RIS). Primary human liver sinusoidal endothelial cells (LSECs) were obtained from 7 explanted human livers at liver transplantation, purified and lines established. The LSECs were incubated in the senescence conditioned media before extraction and isolation of RNA.
