The goal was to investigate RNA toxicity induced by the expansion of CAG triplet repeat sequences and its effects on transcriptome, including mRNAs and small RNAs. SK-N-MC neuroblastoma cells were used as a model in this study. Three conditions were used, including the non-transfected cells, the EGFP-CAG27 transfected cells (with a plasmid construct containing EGFP and 27 consecutive CAG tripleet repeat sequences), and the EGFP-CAG78 transfected cells. In addition, samples of the same three conditions, but treated with a small molecule, DB213, were supplemented. It is recomended to use all samples of the three conditions, in both untreated and treated groups, for performining differential gene expression analysis with the RNA-seq data.
