3D-scanning SAXS on stroke affected mouse brain slices as part of a multi scale approach

Understanding nerve fiber organization is crucial for studying neurodegenerative diseases and stroke recovery. This project employs small-angle X-ray scattering (SAXS), 3D polarized light imaging (3D-PLI), scattered light imaging (SLI), and serial scanning electron microscopy (SSEM) to analyze regenerating fibers in stroke-damaged mouse brain tissue. 3D-scanning SAXS provides essential myelin density data as well as 3D orientation, validating fiber trajectories from 3D-PLI and SLI. Conducted at the ID13 beamline, 3D-scanning SAXS experiments will optimize resolution while minimizing beam damage for subsequent SSEM. Imaging both damaged and intact brain regions will refine imaging methodologies and improve fiber mapping accuracy. This multi-modal approach will enhance our understanding of nerve fiber regeneration and advance neuroimaging techniques.
