In this experiment the binding of SOX2, B-catenin, and LEF1 was assayed using ChIP-seq following differentiation of WT, T/Bra mutant, and Cdx1,2,4 triple mutant embryonic stem cells to caudal ebiblast progenitors. SOX2 binding in epiblast cells was assayed for comparison. Triplicate chromatin immunoprecipitations from independent biological replicates were performed for each factor analyzed in three genetic backgrounds. Binding of each factor was compared between WT caudal epiblast progenitor cells , T/Bra mutant, and Cdx1,2,4 triple mutant progenitors differentiated in the presence of the WNT agonist CHIR99021. SOX2 binding was also compared between caudal epiblast progenitors and epiblast progenitors differentiated in the absence of CHIR99021.
