In this study we hypothesized that trochostatin A (TSA), through its epigenetic activity, may efficiently counteract the effect of cryopreservation stress in pig bone-marrow derived mesenchymal stem cells (MSCs). To verify this hypothesis, we attempted to evaluate changes in MSC gene expression profiles caused by a freezing-thawing procedure and evaluated the result of TSA addition on profile of genes expression in this cells. We additionally performed a quantitative analysis of genes associated with critical biological processes affected either by cryopreservation or TSA administration and evaluated their transcriptional activity in cells harvested at different culture stages including: MSC subjected to cryopreservation, TSA-treated cryopreserved MSC and MSC cultured another 24h after TSA removal.
