In a companion study (PRJEB73474), we determined that 27 chromosome-level Pacbio CLR assemblies of Arabidopsis thaliana accessions are all ∼120 Mb in size. This is noteworthy because we have previously estimated by flow cytometry that A. thaliana genomes vary more than 10% in genome size. Regardless of the sequencing technology, it is still extremely challenging to assemble major satellite repeats. Therefore, we employed k-mer and BLAST-based approaches in PCR-free Illumina short reads to establish the relationship between genome size and three major satellite repeats, namely centromeres, 45S and 5S rDNAs. In this project, we release PCR-free Illumina short-reads of 61 A. thaliana accessions to complement data published in the above-mentioned study.
