This dataset contains spatial transcriptomics data profiling the dorsal hippocampus of one-month-old 5xFAD heterozygous mice and age-matched wild-type littermates. Fresh-frozen brain tissue was cryosectioned at 6 µm and processed using the NanoString GeoMx Mouse Whole Transcriptome Atlas (WTA). Regions of interest (ROIs) were spatially selected based on immunofluorescent staining for neuronal (NeuN) and glial (GFAP, Iba1) markers to specifically isolate and profile the cell body layers of the CA1, CA3, and Dentate Gyrus (DG) subregions. The data files included comprise tabular comma-separated value (.csv) files representing both the raw mRNA transcript counts and normalized gene counts for each subregion and biological replicate. This data was collected to test the hypothesis that synaptic hyperexcitability and subcellular metabolic dysfunction emerge early in the 5xFAD mouse model (1 month of age) and contribute to region-specific neuronal vulnerability before overt plaque burden.
