Human neuroblastoma cell line IMR32 expressing Cas9 was transduced with the Brunello genome-wide gRNA library, followed by co-culturing with primary human lymphocytes +/- nivolumab. After 6 days, cells were harvested and gRNA frequencies were defined using next-generation sequencing. Depleted gRNAs after nivolumab represent resistance genes to nivolumab.
