RePORT-Brazil CD4 T cell objects

Single-cell data generated from RePORT-Brazil Cohort B, which enrolled close contacts of patients with pulomonary tuberculosis. This dataset includes samples from 58 participants (25 who progressed to TB disease over the 24 months after enrollment, and 33 who did not progress in the same period). PBMCs from time of enrollment--at a time when there were no signs/symptoms of disease--were thawed and incubated overnight in three conditions: unstimulated ("Media"), with a pool of 300 synthetic peptides derived from immunodominant Mtb antigens ("MTB300"), or with gamma-irradiated/killed Mtb strain H37Rv ("gRv"). Samples were submitted for Cellular Indexing of Transcriptomes and Epitopes by sequencing (CITE-seq) with VDJ/immune repertoire sequencing. The data were analyzed with Cell Ranger, souporcell for demultiplexing, a multi-algorithm approach for doublet handling, and iterative reclustering to computationally isolate CD4 T cells. Selected T cell receptors (TCRs) from the data were experimentally tested in a reporter assay for recognition of Mtb antigens.
