Diet steatosis in commercial crossbreed swine

This study aims to fully characterize the changes taht undergoes hepatic transcriptome due to dietically induced steatosis in commercial crossbreed pigs Overall design: 8 Landrace x Large White pigs recieved a steatotic diet during two months, sampling took place before and after the intervention. Liver biopsies were immediately removed and frozen in liquid nitrogen. RNA from each liver was isolated using Tri-reagent (Ambion). DNA contaminants were removed by TURBO DNAse treatment using the DNA removal kit from Ambion. RNA was quantified by absorbance at A260/280. The integrity, concentration and purity of RNA was verified by a Biorad experion chip (Hercules, CA, USA) Finally, 4 pools (2 animal samples in each pool) for each condition (initial state (non-steatosis) and final state (steatosis)) were prepared and studied by RNAseq. Results of RNAseq were verified by qPCR.
