The project aimed to study the effect of drought and esca expression on grapevine Ascomycota community using DNA metabarcoding. We collected 251 grapevine samples: 48 petioles, 47 stems of the current year, 39 canes, 117 trunk samples (43 healthy wood, 36 black necrosis, 29 white rot, and 9 brown stripes) at the end of the experiment presented in Bortolami et al. (2021; https://doi.org/10.1073/pnas.2112825118). Plants were mature 30-y-old Vitis vinifera cv. Sauvignon blanc from three treatments: well-watered control asymptomatic and esca symptomatic vines, and asymptomatic vines under water deficit. We realized fungal GTAA amplification (primers GTAA182f and GTAA526r; Morales-Cruz et al. 2018) and Illumina MiSeq sequencing.
