Chromosome-level genome of Berberis wilsoniae

This study conducted whole genome assembly on diploid plant Berberis wilsoniae. We generated the first chromosome-scale reference genome of B. wilsoniae by integrating PacBio HiFi, Hi-C, and Illumina sequencing data. The assembled genome size was 1.23 Gb, with a contig N50 of 76.96 Mb and a scaffold N50 of 87.57 Mb; 99.16% of the assembly was anchored onto 14 chromosomes. The assembly showed high completeness and accuracy, with a BUSCO completeness of 97.5% and a QV of 48.47. A total of 50,808 protein-coding genes were annotated. These results establish a key genomic resource for elucidating berberine-related metabolism in B. wilsoniae and provide new comparative evidence for the origin and diversification of defensive secondary metabolic pathways in Ranunculales and early-diverging eudicots.
