In this study, we identified that the long non-coding RNA actin filament associated protein 1 antisense RNA 1 (AFAP1-AS1), which is highly expressed in lung adenocarcinoma, regulates RNA alternative splicing by recruiting splicing factors such as serine and arginine rich splicing factor 1 and 3 (SRSF1 and SRSF3), promoting their liquid-liquid phase separation. To investigate the effect of AFAP1-AS1 on alternative splicing, we overexpressed and knocked down AFAP1-AS1 in lung adenocarcinoma cell lines and obtained all regulated alternative splicing events through RNA seq, exploring the potential mechanism of AFAP1-AS1 promoting the occurrence and development of lung adenocarcinoma. Overall design: In A549 cells, RNA sequencing was performed on control and experimental groups after overexpression or knockdown of AFAP1-AS1.
