Flag-EMB140 proteins were co-expressed with GFP-UBP5 or GFP proteins in HEK293T cells. These were then incubated with total protein extracts from 30 DAP ubp5-ems#1 kernels, along with the proteasome inhibitor MG132. After 1-hour incubation, Flag-EMB140 was precipitated using Anti-DDDDK-tag (anti-Flag) mAb-Magnetic Agarose beads (CAT#: M185-10, MBL), followed by SDS-PAGE separation for in-gel digestion.
