Mouse arteriovenous fistula model RNAseq

Male C57BL/6J mice aged eight weeks were employed and fed in Huazhong University of Science and Technology’s Institutional Animal Care, and a mouse jugular AVF model was created as previously reported. Briefly,the ligation of the left kidney’s upper pole artery and right nephrectomy were carried out simultaneously to establish CKD in male C57BL/6J mice (6-8 weeks). One month later, after the intraperitoneal injection of pentobarbital sodium (100 mg/kg) to induce anesthesia, an end-to-side anastomosis between the external jugular vein (EJV) and the internal carotid artery (ICA) was performed using an 11-0 silk suture. At postoperative days 3, mice were euthanized with an intraperitoneal injection of pentobarbital sodium (100 mg/kg), and then the contralateral EJVs and the AVF’s venous limbs were collected for RNA sequencing.Male C57BL/6J mice were raised in a particular pathogen-free barrier facility. Similarly, CKD was carried out as previously described four weeks before the infrarenal AVF model creation. Then, a 25-gauge needle is inserted from the aorta into the nearby inferior vena cava (IVC)following anesthesia.A technically successful AVF establishment was verified by the pulsating arterial blood flow in the IVC.The venous outflow veins were gathered on postoperative days 0, 3 for RNA sequencing.
