We examined how Cxcl16 deficiency may alter autoimmune responses targeting pancreatic islets, with a focus on changes in islet-infiltrating CD4 and CD8 T cells. Overall design: Pancreatic islets were isolated and handpicked from 12-week-old female NOD and NOD.Cxcl16-/- mice. The islets were dispersed by a non-enzymatic approach and the dispersed islet cells of either strain were used for FACS-sorting CD45+ immune cells for single-cell RNA-seq analysis.
