2-DG protects mouse from DSS-induced inflammatory bowel disease

Anti-proinflammatory cytokine therapies against interleukin (IL)-6 and tumor necrosis factor (TNF)-a are major advances in treating inflammatory diseases. We found that 2-deoxy-D-glucose (2-DG), a simple monosaccharide, attenuated IL-6 responses by inhibiting N-linked glycosylation of the IL-6 receptor gp130. Aglyco forms of gp130 failed to bind IL-6 and to activate downstream signals. 2-DG completely inhibited dextran sodium sulfate-induced inflammatory bowel disease that was partially dependent on IL-6. To understand the mechanism of DSS-induced colitis with or without 2-DG, we analysed the mRNA expression profiles of mouse colons from DSS-induced colitis with or without 2-DG . The RNeasy Mini Kit (Qiagen) was used to extract total RNA from mouse colons. For microarray, total RNA was extracted from five mock colons, five colons treated with 2% DSS drinking for 7days, five colons injected 10mg 2-DG for 7days,or five colons treated with 2% DSS drinking and injected 10mg 2-DG for 7days . TAKARA BIO (the company for biological technical service) performed the microarray analysis.
