Sequencing results of oligonucleotides libraries

Complex oligonucleotide (oligo) libraries are essential materials for various biotechnological applications. However, purification methods that can be applied to highly complex oligo libraries in a highly parallel manner to lower the error rate of the libraries are lacking. Therefore, we propose a purification method with single-base resolution that can be applied to complex libraries consisting of oligos of different lengths. Both hands-on and a next-generation sequencer based protocols for coupling the intended number of nucleotides to the oligos were demonstrated.
