Direct comparison of distinct naïve pluripotent states in human embryonic stem cells

Several culture conditions have demonstrated successful conversion of primed human embryonic stem cells (hESC) towards a naïve state without pre-ectopic expression of naïve markers, such as naïve human conversion media (NHSM) or reverse toggle protocol (RT) and our published in-house developed naïve conversion media (NCM) as well using minimal growth factors/cytokines. We report here for the first time, a direct comparative analysis of three in-house derived hESCs converted in the aforementioned naïve media in the same lab, thereby providing a more unbiased approach to study the underlying variances between these differentially converted naïve hESCs.
