The soft regions of a thrombus tend to be more susceptible to rt-PA -mediated thrombolysis and are more easily removed by mechanical thrombectomy than the stiff counterpart region. However, little is known about the molecular differences between the soft and stiff regions of human arterial thrombus. A comprehensive molecular pathology of the soft and stiff regions of thrombus will provide new insights into effective therapeutics for thrombosis. Here, we developed a spatial proteomics workflow combining proteomics with laser capture microdissection to analyze 12 fresh human thrombus samples with Masson trichrome staining to identify stiff and soft regions in acute carotid and coronary artery thrombi.
