The analysis of large cohorts of human tumors of different origin (more than 700 tumor samples) showed that CDKN1B mutations are present only in Luminal Breast Cancer (LBC), at low frequency and subclonal levels. The analysis of circulating cell free DNA showed that CDKN1B mutations may be found at higher frequency in metastatic LBC patients. Somatic CDKN1B mutation often resulted in p27kip1 protein truncation. LBC cells engineered to express a comparable number of copies of p27kip1 wild type or truncated proteins showed that deletion of p27kip1 C-terminus led to the loss of p27kip1 oncosuppressive properties.
