Effect of obesity and metformin on subcutaneous Lewis lung carcinoma cell-line tumors in mice

Male, 2-3-month old, C57BL/6J mice were treated with high-fat diet for 16 weeks to induce obesity. Age/sex-matched control mice were fed normal diet. Cohorts from obese and non-obese groups (continued on same diet) were injected subcutaneously (s.c.) with Lewis lung carcinoma (LLC) cells and subsequently monitored until established tumors were noted (~day 11), when mice were injected intra-peritoneally every two days with metformin (50 mg/kg in phosphate-buffered saline [PBS]) or PBS alone. Tumors of mice (n = 5-7 for each of 4 groups) were harvested after 3 weeks of cell injection in mice. Total RNA isolated from frozen tumors after pulverization in liquid nitrogen was subjected to RNA sequencing. Sequencing libraries were prepared using KAPA mRNA HyperPrep kit with 15 PCR cycles (0.5 ug RNA input). Libraries were sequenced on NovaSeq 6000 instrument. Libraries of samples np1-3, nm1-4, op1-3, and om1-2 were sequenced together on same flow cell to obtain single-end 101 b reads. Libraries of other samples were sequenced together in a different flow cell (separate sequencing run) to obtain paired 101 b reads. Demultiplexed fastq data are provided here; the data are adapter-trimmed with bcl2fastq 2.20 (90% identity threshold). Data from two mouse experiments are provided.
