Genetic and pharmacological evidence for kinetic competition between alternative poly(A) sites in yeast - cordycepin treatment

PAT-seq approach was used to determine changes to 3'UTR length in yeast upon addition of 20µg/ml cordycepin (3'deoxyadenosine) for 0, 5, 10, 20 or 40 minutes BY4741 yeast cells were grown at 30ºC until an OD600 of 0.6 was reached then 20µg/ml cordycepin was added for 0, 5, 10, 20 or 40 minutes before harvesting. Samples were analysed via PAT-seq 3'UTR focused deep-sequencing in replicate using the Illumina Hiseq1500 platform.
