Messenger RNA (mRNA) regulation at the transcript level represents an important layer of control in cellular senescence. To investigate how NAT10-mediated N4-acetylcytidine (ac4C) modification influences transcript dynamics in senescent colonic epithelial cells, we performed transcriptome-wide RNA sequencing following transcriptional arrest using actinomycin D (ActD).Senescence was induced in human colonic epithelial cells, after which cells were transfected with either control siRNA (NC-Si) or NAT10-targeting siRNA (NAT10-Si). Transcription was subsequently halted by ActD treatment, and total RNA was collected at 0 h, 2 h, and 4 h post-treatment. RNA-seq was conducted to quantify transcript abundance over time and to characterize global transcript-level changes associated with NAT10 knockdown in the senescent context.This dataset enables systematic evaluation of NAT10-dependent regulation of transcript dynamics during colonic epithelial senescence and provides a resource for identifying transcripts whose temporal expression patterns are altered upon disruption of ac4C modification.
