During embryo implantation, endometrial epithelial cell is the first cell type to contact the embryo. These cells undergo a series of cellular and molecular changes to establish uterine receptivity for embryo implantation. This process is mainly regulated by estrogen and progesterone. Low estrogen levels decide to open the implantation window in the uterus, while high estrogen levels can close the implantation window and impair endometrial receptivity. Our previous study showed that supraphysiological estrogen level produced during superovulation lead to embryo implantation failure, accompanied by a significant increased the mRNA expression of proline-rich acidic protein 1 (Prap1). PRAP1 is a secreted protein in epithelial cells of the gastrointestinal tract, uterus, liver, and kidneys. PRAP1 is abundantly expressed in the uterus in late pregnancy and disappears from the uterus within 3 days after birth, previously named pregnancy-specific uterine protein (PSUP). PRAP1 is involved in cancer responses by inducing cell cycle arrest, affecting cell migration ability and influencing the immune microenvironment, playing a crucial role in maintaining the epithelial cells homeostasis. It is also involved in multiple cellular damage responses, cellular stress, and repair-oriented targets as a cell protective factor. The aim of this study was to investigate whether overexpression of Prap1 in mouse endometrial epithelial cells affects embryo implantation.
