LSD1/KDM1A Maintains Genome-wide Homeostasis of Transcriptional Enhancers [GRO-Seq]

We performed GRO-Seq in Lsd1-WT and Lsd1-GT mouse embryonic stem cells (mES).SMCX_WT (KDM5c_WT) and SMCX_KO (KDM5c_KO) represent two additional mES cell lines used in the GRO-Seq study. We performed GRO run on mES cells in technical duplicates. Nuclei isolation and GRO was conducted as described in PMID:19056941 and PMID:21572438 with minor modifications. In addition to the presence of 0.2% IGEPAL CA-630, GRO on Kdm5c mES and Lsd1 mES were done in presence of 0.5% and 0.25% N-Lauroylsarcosine, respectively for 8 min at 30 degrees C.
