Quantitative secretome atlas of soluble E-cadherin-driven vascular leakage in the lung pre-metastatic niche of salivary adenoid cystic carcinoma

This data set reports the comprehensive, label-free quantitative proteomic profile of conditioned media from high-metastatic (SACC-LM) versus low-metastatic (SACC-83) salivary adenoid cystic carcinoma cells. LC-MS/MS analysis identified and quantified 5,342 proteins (FDR ≤ 1 % at PSM, protein level), of which 1,847 were significantly differentially secreted (≥ 2-fold change, p ≤ 0.05). Bioinformatic enrichment highlighted over-representation of cell-adhesion molecules, serine hydrolases and apical-junction components, with soluble E-cadherin (sE-cad/CDH1) displaying the strongest up-regulation (22-fold) in the metastatic secretome. The inventory provides a resource for investigating tumour-derived factors that destabilise endothelial tight-junctions via the Rac2/LIMK/cofilin-1 axis and offers candidate biomarkers for monitoring or targeting the lung pre-metastatic niche in SACC.
