To investigate the role of AGO10 in the buffering of cytokinin responses we grew the ago10-1 mutant and the Col-0 wildtype on plates for six daysbefore treting with mock solution or cytokinin for 2 hours. Then root tips excised and RNA was extracted using a commercial kit.
